despiteは、「de＝下」＋「spite＝spect＝見る」から出来上がっている単語で、「下に見る」－「見下す」－「軽蔑する」の意味を表します。despiteに当たる日本語は「～をよそに」という表現です。「よそに」は「他所」とか「余所に」と書いて、気持ちがここになく他所にあること、つまり「無視して」くらいの意味です。
■両親は心配していたのだが、その心配をよそに、彼は放蕩の限りを尽くした。
Despite his parents’ concern, he lived an extravagant life.

ほら、立派な逆接・譲歩になってるでしょ！in despite of Aの省略形としてdespite Aやin spite of Aが出来上がりました。ちなみにinは「状況のin」、A of Bで「ＢをＡする」の目的格です。だから「Ａを軽視した状態で」が原義です。２つ以上の語が集まって１つの前置詞として機能するものを「群前置詞」と言いました。この３つは全部前置詞です。
だから、直後には名詞が来てますね。
